                    The Study of Vetiveria zizanioides (L.) Nash Using Temporary Immersion Bioreactor as

                               a Raw Material for the Root–knot Nematode Control Products

                                                          Abstract

                          The  objective  of  this  research was  to study the  appropriate  factors  for the
                   production of  Vetiveria zizanioides (L.)༛Nash༛by༛using  Temporary Immersion Bioreactor

                   (TIB).༛This༛studies༛were༛divided༛to༛the༛micropropagation༛culture༛as༛follows:
                          For the initiation stage from axillary bud or V. zizanioides (L.)༛Nash༛natural༛seedling༛

                   was༛highly༛contaminated༛with༛fungi༛and༛bacteria.༛Therefore,༛2༛mg/l༛PPM༛should༛be༛used༛

                   into༛modified༛MS༛(1962)༛medium༛supplement༛with༛1-2༛mg/l༛BA༛and༛0.1༛mg/l༛NAA༛to༛reduce༛
                   the contamination༛and༛increase༛sterile༛cultured.

                          For༛the༛multiplication༛stage,༛the༛related༛factor༛was༛growth༛regulators,༛1-2༛mg/l༛BA༛
                   and༛0.1༛mg/l༛NAA༛were༛supplemented༛in༛modified༛MS༛(1962)༛liquid༛medium༛for༛TIB༛by༛

                   feeding every 4 hours and about 5 minutes each time in༛total༛for༛6༛time༛a༛day.༛By༛the༛way༛

                   the༛culturing༛of༛miniature༛explant༛(<0.5༛cm༛height༛and༛5༛shoots༛per༛clump)༛were༛cultured༛
                   40༛clump༛in༛700༛ml༛glass༛vessel༛which༛was༛contained༛modified༛MS༛(1962)༛liquid༛medium༛

                   supplement༛ with༛ 1༛ mg/l༛ BA༛ and༛ 0.1༛ mg/l༛ NAA,༛ the༛ multiplication༛ was༛ 518.8༛

                   shoots/container.༛Likewise༛the༛higher༛explant༛(0.6-2.0༛and༛2.1-6.0༛cm༛height)༛should༛be༛
                   cultured༛30༛clump༛in༛the༛modified༛MS༛(1962)༛liquid༛medium༛supplement༛with༛2༛mg/l༛BA༛

                   and༛0.1༛mg/l༛NAA,༛the༛multiplication༛was༛686.1༛and༛389.1༛shoots/container,༛respectively༛

                   within༛4༛weeks.
                          For༛the༛elongation༛and༛rooting༛stage,༛NAA༛should༛be༛used༛in༛order༛to༛induce༛root༛

                   initiation due to NAA stimulated more roots than IBA at the same concentration.༛By means
                   of༛0.1༛mg/l༛NAA༛gave༛the༛highest༛number༛of༛roots,༛moreover,༛the༛highest༛number༛of༛shoots༛

                   per clump༛were༛25.63༛roots༛and༛13.53༛shoots༛per༛clump,༛respectively༛within༛2༛weeks.༛The༛
                   plantlets were༛transplanted༛with༛100%༛survival༛percentage.༛It༛was༛not༛different༛in༛0.5༛mg/l༛

                   NAA  but  when cultured  for༛3༛weeks༛and༛then༛transplanted,༛the༛number༛of༛new༛roots༛

                   increased༛to༛19.43༛roots༛per༛clump༛within༛2༛weeks༛after༛transplanting.
                          According to the appropriate culturing for the bioactive compound of V. zizanioides

                   (L.)༛Nash༛by༛using༛TIB,༛it༛was༛found༛that༛the༛culture༛period༛of༛30-45༛days༛was༛most༛likely༛

                   to༛ give༛ the༛ highest༛ Total༛ Phenolic༛ Content༛ (TPC).༛ The༛ concentration༛ of༛ 1.0༛ mg/l༛ BA༛
                   provided more TPC than low concentration BA and without growth regulators (controls)

                   were༛more༛likely༛to༛provide༛TPC༛than༛0.1༛mg/l༛NAA༛and༛0.25༛mg/l༛BA,༛respectively.༛In༛
